A deoxyribonucleic acid polymerase I-deficient mutant of Bacillus subtilis.
نویسندگان
چکیده
A methyl methanesulfonate sensitive mutant of Bacillus subtilis appears to lack the enzyme DNA polymerase I. The absence of polymerase I permits the detection of a second DNA polymerizing enzyme in the mutant. This second polymerase is inhibitable by sulfhydryl antagonists, and shows maximal activity on double-stranded DNA substrates with many single strand breaks. Poly[d(A-T)] is less efficiently used as a substrate, in contrast to the substrate preferences of polymerase I. Compared to the wild type, the mutant is more sensitive to ultraviolet and x-irradiation, and is inefficient in reactivation of ultraviolet-irradiated SPOl bacteriophage, but is normal in genetic recombination.

متن کاملIn vitro repair of x-irradiated DNA extracted from Bacillus subtilis deficient in polymerase I.
DNA extracted from x-irradiated cells of a mutant of Bacillus subtilis deficient in DNA polymerase I has greatly reduced biological activity. This DNA, which has many single-strand nicks, can be repaired in vitro with purified DNA polymerase I and DNA ligase or with lysates of wild-type B. subtilis cells, which restore biological activity and increase the single-strand molecular weight. Lysates...
متن کاملInhibition of iron uptake and deoxyribonucleic acid synthesis by Desferal in a mutant strain of Bacillus subtilis.
In the Bacillus subtilis mutant 1D-4, the hydroxamate Desferal inhibited growth, iron uptake, and deoxyribonucleic acid synthesis but did not quantitatively affect synthesis of ribonucleic acid and protein.
متن کاملLipiarmycin-resistant ribonucleic acid polymerase mutants of Bacillus subtilis.
Lipiarmycin inhibited the activity of deoxyribonucleic acid-dependent ribonucleic acid (RNA) polymerase in vitro. We showed that inhibition was due to interference by lipiarmycin with the activity of sigma-containing molecules of RNA polymerase. Transcription by core enzyme was relatively resistant to the drug, but addition of sigma led to highly drug-sensitive RNA synthesis. We isolated lipiar...
متن کاملRegulation of the bacterial cell wall: analysis of a mutant of Bacillus subtilis defective in biosynthesis of teichoic acid.
Bacillus subtilis 168ts-200B is a temperature-sensitive mutant of B. subtilis 168 which grows as rods at 30 C but as irregular spheres at 45 C. Growth at the nonpermissive temperature resulted in a deficiency of teichoic acid in the cell wall. A decrease in teichoic acid synthesis coupled with the rapid turnover of this polymer led to a progressive loss until less than 20% of the level found in...
